Oxidative Stress in Diabetes Mellitus and the Role Of Vitamins with Antioxidant Actions

Type 2 diabetes mellitus is a leading cause of morbidity and mortality worldwide, given its serious associated complications. Despite constant efforts and intensive research, an effective, ubiquitous treatment still eludes the scientific community. In this sense, we revisited the most promising therapeutic interventions both non-pharmacological and antidiabetic drugs targeting oxidative stress and inflammation to hinder progression of vascular complications of diabetes. This review article gives particular attention to the relevance of mitochondrial function, an often ignored and understudied organelle in the vascular endothelium. We highlight the importance of mitochondrial function and number homeostasis in diabetic conditions and discuss the work conducted to address the aforementioned issue by the use of various therapeutic strategies.

Oxidative Stress in the Pathogenesis/Treatment of Diabetes and its Complications

Diabetes: Oxidative Stress and Dietary Antioxidants, Second Edition, builds on the success of the first edition, covering updated research on the science of oxidative stress in diabetes and the potentially therapeutic usage of natural antioxidants in the diet and food matrix. The processes within the science of oxidative stress are not described in isolation, but rather in concert with other processes, such as apoptosis, cell signaling and receptor mediated responses. This approach recognizes that diseases are often multifactorial and oxidative stress is a single component of this. Since the publication of the first edition, the science of oxidative stress and free radical biology continues to rapidly advance with thousands of the research articles on the topic. New sections in this update cover the role of dietary advanced glycation end products AGEs in causing OS in diabetes, oxidative stress and diabetes-induced bone metabolism, and oxidative stress and diabetic foot ulcer.

A Review on Oxidative Stress, Diabetic Complications, and the Roles of Honey Polyphenols

Oxidative stress is increased in metabolic syndrome and type 2 diabetes mellitus T2DM and this appears to underlie the development of cardiovascular disease, T2DM and diabetic complications. Core tip: Oxidative stress is underling in the development of cardiovascular disease, type 2 diabetes mellitus T2DM and diabetic complications. Aerobic life uses oxygen to oxidize metabolism food substrates carbon- and hydrogen-rich to obtain the heat energy and chemical essential for life. When we oxidize molecules with oxygen, the oxygen molecule itself becomes reduced and forms intermediates. In eukaryotic cells, reactive oxygen species ROS are always produced as the consequence of regular physiological metabolism [ 1 ].

Hyperglycaemia, a characteristic of type 2 diabetes mellitus T2DM , substantially leads to the generation of reactive oxygen species ROS , triggering oxidative stress as well as numerous cellular and molecular modifications such as mitochondrial dysfunction affecting normal physiological functions in the body. In mitochondrial-mediated processes, oxidative pathways play an important role, although the responsible molecular mechanisms remain unclear. The impaired mitochondrial function is evidenced by insulin insensitivity in various cell types. Dietary factors are known to influence diabetes, and many natural dietary factors have been studied to improve diabetes. Honey is primarily rich in carbohydrates and is also abundant in flavonoids and phenolic acids; thus, it is a promising therapeutic antioxidant for various disorders.

Oxidative stress plays a pivotal role in the development of diabetes complications, In addition, diabetic cardiomyopathy is a major diabetic complication. Molecular Mechanisms of Glucose Fluctuations on Diabetic Complications

Author s : Ludovica Piconi , Michael A. DOI : In diabetes oxidative stress plays a key role in the pathogenesis of vascular complications, and an early step of such damage is considered the development of an endothelial dysfunction. Hyperglycemia directly promotes an endothelial dysfunction inducing process of overproduction of superoxide and consequently peroxynitrite that damages DNA and activates the nuclear enzyme poly ADP-ribose polymerase. New, low molecular mass compounds that act as SOD or catalase mimetics or L-propionyl-carnitine and lipoic acid, that work as intracellular superoxide scavengers, improving mitochondrial function and reducing DNA damage, may be good candidates for such strategy, and preliminary studies support this hypothesis.

Oxidative stress is suggested to be the crucial factor in diabetes mellitus type 2 DM2 pathogenesis and in the development of diabetic complications. Patients with DM2 may be more susceptible to infections due to hyperglycaemia-induced virulence of various microorganisms. Blood and saliva were collected from patients with DM2.

Accumulating evidence indicates the occurrence and development of diabetic complications relates to not only constant high plasma glucose, but also glucose fluctuations which affect various kinds of molecular mechanisms in various target cells and tissues.

Oxidative Stress in Diabetes Mellitus and the Role Of Vitamins with Antioxidant Actions

Diabetes mellitus is a group of metabolic diseases characterized by hyperglycemia resulting from defects of insulin action, insulin secretion or both [ 1 ]. Diabetes has taken place as one of the most important diseases worldwide, reaching epidemic proportions. Hyperglycemia in the course of diabetes usually leads to the development of microvascular complications, and diabetic patients are more prone to accelerated atherosclerotic macrovascular disease. These complications account for premature mortality and most of the social and economical burden in the long term of diabetes [ 3 ]. Increasing evidence suggests that oxidative stress plays a role in the pathogenesis of diabetes mellitus and its complications [ 4 ].
